ABSTRACT

AlM: To explore the roles of neuronal axon-guidance molecules Slit3 and Robo4 receptor in corneal neovascularization ( CNV ) by study their expression in neovascularized cornea of rats. METHODS: CNV models were established by implantation pellets containing basic fibroblast growth factor ( bFGF ) into corneal stroma. CNV models were measured by biomicroscopy photography. lmmunohistochemical staining and imaging analysis system were used to detect the expression of Slit3 and Robo4 in the models after 1, 4, 7, 10 and 14d. RESULTS:The area of CNV and the expression of Slit3, Robo4 were increased in CNV models compared to that in normal cornea and reached highest level on 7d. And the expression level of Slit3 and Robo4 were significantly correlated with the size of CNV on every time point except 1d (r=0. 84-0. 91, all P CONCLUSlON: The expression of Slit3 and Robo4 may be related to the CNV development. They are potential therapeutical target for CNV.

ABSTRACT

OBJECTIVETo observe the effect of Polygonatum sibiricum on Yin deficiency model rats induced by long-term overload swimming.

METHODExcept for the normal group, all of the remaining rats performed the long-term overload swimming for eight weeks, with five days every week and once every day, to establish the Yin deficiency model. The daily swimming time increased from 10 min to 180 min at the end of the 7th week, with the water depth of 60 cm and the water temperature at 30 degrees C. After the success of the modeling, the rats were orally administered with different doses of aqueous extracts from P. sibiricum (2.5, 10 g x kg(-1)) for eight weeks. After the final administration, their blood were collected from orbits to measure immunoglobulin A, G and M (IgA, IgG, IgM), interleukin 2 and 6 (IL-2, IL-6) and cAMP, cGMP contents in plasma General behavioral indicators (weight, facial temperature, pain threshold and holding power) of rats were observed during the drug administration.

RESULTCompared with the model control group, aqueous extracts from P. sibiricum was given for eight weeks to significantly increase the rat weight and holding power of Yin deficiency model rats, decrease the facial temperature and the sensitivity of pain threshold, and increase IgA, IgG, IgM and IL-6 content and IgG content in serum, but without statistical difference. Aqueous extracts from P. sibiricum (10 g x kg(-1)) could also increase IL-2 content in serum, and decrease cAMP content and cAMP/cGMP ratio.

CONCLUSIONP. sibiricum could improve the general behavioral indicators (weight, holding power, pain threshold and facial temperature), immunologic functions (IgA, IgG, IgM) and cyclic nucleotide (cAMP, cAMP/cGMP), so as to ameliorate such Yin deficiency symptoms as dysphoria in chestpalms-soles, weight loss, soreness and weakness of waist and knees, immunologic dysfunction and cyclic nucleotide system disorders.

ABSTRACT

AIM: To measure the anatomical character of nasolacrimal dust by spiral CT 3D reconstruction in children.METHODS: The length of nasolacrimal dust and the angles between nasolacrimal dust and vertical plane, horizontal plane and coronal plane in 27 children(54 eyes) and 15 adults(30 eyes) were surveyed using spiral CT 3D reconstruction. While the length of nasolacrimal dust in 7 dead children(14 eyes) body were surveyed as comparison by anatomical method. The RESULTS:were analyzed by software SPSS 13.0 statistically. RESULTS: The length of nasolacrimal dust was 10.06±0.29mm in children and 11.51±1.54mm in adults by spiral CT 3D reconstruction,as 9.95±0.31mm in dead children body by anatomical method, with significant statistical difference between that of children and adults. The angles between nasolacrimal dust and vertical plane, horizontal plane and coronal plane in children were 7.96°±1.62°,73.24°±6.75°,and 12.31°±2.03° respectively, while the corresponding angels in adults were 8.08°±0.63°,72.69°±3.85° and 12.09°±1.21°. The difference between them had no statistical meaning. CONCLUSION: The anatomical data of children nasolacrimal dust obtained from spiral CT 3D reconstruction have important guidance to the therapy of nasolacrimal dust diseases in children.

ABSTRACT

AIM: To study the effects of resveratrol on the proliferation of human retinal vascular endothelial cells induced by cobalt chloride-simulated hypoxia in vitro.METHODS: CoCl2 (100μmol/L) was used to simulate hypoxic condition, and human retinal vascular endothelial cells were cultured in vitro as model. The cell proliferation was determined by MTT method; SABC method was employed to test the expression of vascular endothelial growth factor (VEGF); and computer image analyzer was used to process data. The effects of resveratrol on the proliferation of vascular endothelial cells were observed.RESULTS: Resveratrol inhibited the proliferation of human retinal vascular endothelial cells induced by CoCl2 in a dose- and time-dependent manner in vitro, meanwhile VEGF expression in all groups which were administered medicine was down-regulated. Both kinds of inhibitive effects of resveratrol were statistically significant (P<0.01).CONCLUSION: Resveratrol can significantly inhibit the proliferation of human retinal vascular endothelial cells and the expression of VEGF, which may provide a new approach for prevention and treatment of retinal neovascular diseases.
